Super Quick Ligation

Use spreadsheet to calculate volumes

  1. Mix vector, insert, and buffer 2.
  2. Add water and buffer one and mix.
  3. Add 1ul of T4 Ligase and let sit for 5-30 min.
  4. Mix ligation product w/ cells (DH5-alpha)
  5. Place on ice for 15 minutes
  6. Heat Shock at 42C for 90 seconds.
  7. Place on ice for 2 min.
  8. Add 1/2 ml LB.
  9. Grow in Screw-Cap Tube for 1 hour.
  10. Plate (on Amp+ plates)
